Western blot analysis was performed on whole cell extracts (30 µg lysate) of U-937 (Lane 1), Caco-2 (Lane 2), MCF7 (Lane 3), Jurkat (Lane 4) and LNCaP (Lane 5). The blot was probed with Anti-TRADD Recombinant Rabbit Monoclonal Antibody (Product # 703356, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution). A ~34 kDa band corresponding to TRADD was observed in all the cell lines tested.

The blot was probed with Anti-TRADD Recombinant Rabbit Monoclonal Antibody (Product # 703356, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution). Densitometric analysis of this Western blot is shown in the histogram (Fig b). Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to TRADD.
